I"m working my way through the ACL video, but wondered if someone could explain the "configure" option in the Article Manager. For example, if I go to the Article Manager, options, permissions, and then click on "manager," I notice that a manager can't configure. But I don't understand what "configuring" means, and the yellow pop-up box in Joomla is no help! I think I need to make someone a manager, but am unsure if they'll need to use the "configure" feature.

The configure section in the Article Manager is for setting default configuration for the site. Like whether the author name or date is supposed to show and other details of the the display.

Ususally that's a site-wide configuration and only super-admins need deal with it. Your editors and publishers won't need to deal with the article configuration unless there is some special reason you want them to override the defaults.
